MET Amplification by FISH

Background: c-Met amplification activates downstream pathways of the mitogen-activated protein kinase (MAPK) pathway and is a therapeutic target in NSCLC.

Methodology: The MET probe is labeled in ORANGE and detects copy numbers of the 7q31.2 region. The control probe is labeled in GREEN located in the centromere region of chromosome 7. When the probe is hybridized to a normal cell it will show two orange and two green signals. A single 4-5µM FFPE section with marked tumor area is pretreated and then hybridized to the MET/CEP7 probe spanning ~ 440kb of MET gene located at 7q31.2. MET amplification is identified by an absolute MET copy number > 5.0 and/ or ratio of MET/CEP >2.0 based on validation studies. Signal configurations in a total of 50 tumor cells are enumerated.
